Film
In 1983, Samrat Mukerji made his acting debut as a child artist in the film Karate. In 1996, he acted in the film Ram Aur Shyam in which he played the role of Ram.
In 1997, he appeared as Akbar in the Bollywood film Bhai Bhai alongside Shakti Kapoor.
In 1998, he played the role of Sudhakar in the film Zanjeer. In the film, he acted alongside Aditya Pancholi.
In 1999, he played the role of Balraam in the film Sikandar Sadak Ka.
In 1999, in the film Sar Ankhon Par, Samrat Mukerji acted alongside the renowned Bollywood actor Shah Rukh Khan.
In the same year, he was offered a role in a Bengali film, Tomake Solam.

In 2015, he was once again given the role of Mahisasur in the Bengali TV serial titled Mahisasurmardini. In 2018, Samrat was offered a role in the Bengali TV show Jayang Dehi. The show was aired on Colors Bangla in 2018. From 2020 to 2022, in the Bengali TV serial Gangaram, Samrat Mukerji played the role of Samrat (Sammy) Kumar.

In his acting career, Samrat Mukerji has been signed up for several Bollywood films, which were ultimately shelved by the producers themselves. In 1996, he was offered roles in four Bollywood films, Namak Halal, Ujala, Rankshetra, and Humrahi, which were shelved. In 2002, in the film Shaheed Bhagat Singh, he was offered the titular role of the freedom fighter, Bhagat Singh. The film was shelved by the producer in 2002.
